Isolation und Kultur von Blattprotoplasten von Brokkoli (Brassica oleracea var. italica)

The method for the isolation of leaf protoplasts of broccoli was optimized for the cultivars: Atlantik, Corvet, Futura, Green Vilient, Green Charger, Juwaprim, Neptune and Skiff. Protoplasts had a high vitality when prepared in an enzyme solution containing 1 % cellulysin and 4 % (v/v) pectinase. High enrichment and better survival of the protoplasts was achieved by floatation on 0,5 M sucrose solution. Leaf protoplasts of the cultivars Futura, Neptune and Skiff regenerated into callus. A large portion of the calli developed roots. After further cultivation also somatic embryos could be obtained.
