Characterization of HEV-1 Sar55 Replication in Caco-2 Cells

We have recently shown that the colonderived cell line Caco-2 supports the generation and propagation of hepatitis E virus genotype 1 strain Sar55 (HEV-1Sar55), whereby HEV-1Sar55 replicated to higher titers in comparison to HEV-347832mc. Objective The aim of this study was to further characterize the replication of HEV-1Sar55 in Caco-2 cells. Methods HEV-1Sar55 and HEV-347832mc were generated by transfection of in vitro-transcribed and -capped RNA genomes into Caco-2 cells. Culture supernatant-derived virus from transfected or infected cells was used to infect Caco-2 cells. HEV RNA titers in culture supernatants over time as well as the number of HEV particles that attached to or entered Caco-2 cells were detected by RT-qPCR.
