Serodiagnosis of influenza virus and Newcastle disease virus

Es wurden mehrere rekombinante Proteine des Influenza-A-Virus und des ND-Virus in einem bakteriellen Expressionssystem produziert und durch weitere In-vitro-Verfahren authentisch gefaltet. Die rekombinanten Proteine wurden als Antigene in indirekten ELISA-Assays oder in sogenannten Luminex-Bead “liquid arrays” evaluiert. Es konnte gezeigt werden, dass serologische Assays auf Basis dieser rekombinanten Proteine eine mögliche alternative Methode zu den klassischen, jedoch schwer standardisierbaren, Hemagglutinationsinhibitionsverfahren sind.

New serodiagnostic formats for the detection of influenza virus and NDV infections were established and validated. Here, several recombinant proteins of influenza A virus and ND virus were expressed via bacterial expression systems. Authentic folding of these proteins was achieved in vitro. These recombinant proteins served as antigens in indirect ELISA assays or in Luminex bead-based liquid arrays. It is demonstrated that serological assays based on these recombinant proteins are potential alternative methods to classical yet difficult to standardize hemagglutination inhibition assays.
