Die Trennung von Schweineblutzellen mit Hilfe von Ficoll‐Paque
Nach Ficoll-Paquebehandlung von Schweineblutzellen waren in der oberen Zellfraktion 2,7–18,4% der Ausgangszellkonzentration mit 78,2–94,2% Lymphozyten (Ly), 0–7,5% polymorphkernige Leukozyten (PMNL) sowie 3,3–13,3% Monozyten (Mono) und in der Bodenfraktion 61–90% der Ausgangszellkonzentration mit 11,1–43,6% Ly, 32,8–88,9% PMNL und 0–0,8% Mono enthalten. Eine zweite Reinigung der Bodenfraktion ergab in der oberen Zellfraktion nur 1–3% der Ausgangszellzahl, während 55 bis 65% der ursprünglich vorhandenen Zellen in der Bodenfraktion vorhanden waren, die sich aus 11,2–51,5% Ly, 32,8–88,9% PMNL und 0–0,9% Mono zusammensetzte. Durch Zentrifugierung durch Ficoll-Metrizoatgemische verschiedener Dichte gelang keine weitergehendere Trennung von Ly und PMNL. Somit besitzt ein erheblicher Teil der Schweine-Ly offensichtlich eine ähnliche Dichte wie die PMNL und sedimentieren deshalb mit diesen.
After treatment of porcine blood cells with Ficoll-paque 2.7–18.5% of the original cell concentration with 78.2–94.2% lymphocytes (Ly), 0–7.5% polymorphonuclear leukocytes (PMNL) and 3.3–13.3% monocytes (Mono) were isolated in the upper cell layer, and 61–90% of the original cell concentration with 11.1–43.6% Ly, 32.8–88.9% PMNL and 0–0.8% Mono were isolated in the bottom layer. After a second Ficoll-paque treatment of the latter only 1–3% of the initial cell number were in the upper layer and 55–65% of the cells in the bottom layer, which contained 11.2–51.5% Ly, 32.8–88.9% PMNL and 0–0.9% Mono. No further separation of Ly and PMNL was achieved by centrifugation through Ficoll-metrizoat mixtures of various density. Thus, a considerable proportion of the pig Ly apparently possess a density similar to that of the PMNL and therefore sediment together with them.
