Neue Wege in der Rinderzüchtung: Somatisches Klonen und transgene Rinder

Das somatische Klonen ist bis heute bei mehr als einem Dutzend Spezies erfolgreich gewesen. Die Erfolgsraten, d.h. der Anteil geborener Nachkommen aus dem Klonprozess, sind niedriger im Vergleich zu konventionell produzierten Tieren. Die weltweit intensive Forschung hat jedoch zu deutlichen Verbesserungen geführt, so dass das Klonen heute bereits bei Rind und Schwein für spezifische Fragestellungen in Forschung und Anwendung eingesetzt werden kann. Das Klonen bietet ein überaus großes Anwendungspotential, insbesondere in der Grundlagenforschung, als therapeutisches Klonen in der Biomedizin sowie als reproduktives Klonen. Für die Tierzucht sind insbesondere die verbesserten Möglichkeiten zur Produktion transgener Tiere von Bedeutung. Die Effizienz des additiven Gentransfers kann erheblich erhöht werden gegenüber dem bisher üblichen Mikroinjektionsverfahren. Auch die selektive Ausschaltung eines Gens (Knockout) ist erstmals durch das Klonen beim Nutztier möglich. Ein weiteres wichtiges Anwendungsfeld ist die Erhaltung genetischer Vielfalt durch Einsatz somatischer Zellen von Tieren bedrohter Rassen. Die Anwendungsperspektiven des somatischen Klonens könnten durch die Verfügbarkeit pluripotenter Stammzellen deutlich erweitert werden. Die neuen genomischen Techniken und das somatische Klonen unter Einbeziehung pluripotenter Stammzellen können wesentlich dazu beitragen, eine effiziente, diversifizierte, zielgenaue und damit nachhaltige Tierproduktion zu entwickeln.

Somatic cloning has been successful in more than a dozen mammalian species. Success rates are still lower than with conventional reproduction or application of other biotechnologies such as embryo transfer or in vitro embryo production. However, recently significant progress has been made which allows to use porcine and bovine cloning in specific application fields in research and animal breeding. Somatic cloning bears great application potential, mainly in three areas: Basic research, therapeutic cloning in biomedicine and reproductive cloning. With the aid of somatic cloning, transgenic animal production can be significantly improved. Additive gene transfer is much more efficient than the previously used microinjection technology. The use of somatic cloning allows for the first time to specifically delete genes (knockout) as has been shown for the bovine prion locus. The application potential of somatic cloning could be significantly expanded by the availability of pluripotent stem cells. A novel promising approach is the production of induced pluripotent stem cells (iPS) by reprogramming somatic cells via over-expression of only 4 pluripotency related genes. Another important area of application is maintenance of genetic diversity by cryopreservation of somatic cells from endangered species/breeds and their reconstruction by somatic cloning. The novel genomic technologies and somatic cloning based on the use of pluripotent stem cells will make a significant contribution towards development of an efficient, diversified and targeted and thus sustainable animal production.
